For structural work on any given protein, having access to multiple crystal forms is extremely powerful; for example, different crystal forms can show different conformational states, and structure support for lead optimisation requires a packing arrangement that allows ligands to bind.
Once a protein or protein construct is known to crystallize, multiple approaches can be applied to modulate the sample, including surface mutation, changing construct length, chemical modification of surface residues, or extensive exploration of crystallization chemistry.
